Closing a peptide into a ring changes what the molecule can reach. This review sets out how macrocyclic peptides are built and selected — phage display, genetic code expansion with noncanonical amino acids, and a range of cyclization chemistries — and what constraining the backbone yields: target affinity, metabolic stability and pharmacokinetic behavior that sit between small molecules and biologics.
It surveys the pharmacological activities reported for the class and describes where candidates currently stand in development, then treats the same molecules as laboratory reagents rather than only as candidates: selective probes for modulating target proteins, and high-affinity ligands for biomolecular imaging.

All submissions undergo initial editorial screening. Manuscripts that meet the journal’s scope and minimum requirements proceed to independent peer review. Single-blind by default; double-blind review is available on request. Manuscripts that proceed to external peer review are normally evaluated by at least two independent subject-matter experts.
Editorial decisions are based on scope, scientific quality, methodological rigor, ethical compliance, reporting quality, and relevance to the journal. Evidence is weighed against the claims a manuscript actually makes, which is set out on the Aims and Scope page.
The journal is open access. Articles are published under the license stated on each article, and an article processing charge applies to accepted manuscripts; the current terms are on the Article Processing Charges page.
